Donald Love's business model requires a gang war, and a gang war requires the Yakuza to believe the Cartel killed Kenji. So Claude takes a Colombian car to the Newport multistorey where Kenji is waiting for a meeting, and runs him over in it.
It is the mission that ends the second act and the one that costs the most if taken early: everything Kenji has left to give disappears the moment it is finished.
This closes his strand permanently. Six of his jobs become unreachable, which is the largest single block of missions GTA III will take away from a player who is not paying attention.
The frame depends on being seen as a Colombian. Getting out of the Cruiser inside the car park, or during the escape, and being spotted by the Yakuza fails the mission.
The Yakuza in the multistorey can be thinned out from a lower level with a launcher bought at Phil's before going up for Kenji himself, and the corner ramp is the fast way back down.
Somebody has to survive to report what they saw, or there is nobody to blame the Cartel. Then the car has to be dumped.

Left for dead by his partner during a bank job and freed from a prison transport by an explosion he had nothing to do with. Says nothing at all for the length of the story and works his way through every organisation in Liberty City regardless.
